The Millers head into tonight's clash looking to make it five wins out of five for the new season, and to claim their second Championship scalp of the competition after knocking out Wednesday in round one.
They go into the game on the back of a 3-1 win over Chester City at the weekend, but still have major fitness doubts over captain Pablo Mills and striker Ryan Taylor, who have been laid low with a virus.
Defender Marc Joseph may return but is still struggling, Drewe Broughton is expected to continue to lead the line following a decent debut outing.
Wolves, who came from behind to dispose of Accrington in the first round, have to do without George Elokobi and Lewis Gobern. Goalkeeper Matt Murray is still a long term injury victim.
The visitors are expected to rest a few of their bigger name players and the Millers will be hoping to take full advantage, Mark Robins explained to Radio Sheffield: "Wolves are very good side and I expect them to be in the shake up for promotion this season.
"They are quick, powerful, aggressive and play good football. It will a take a big effort from us to beat them but we know we are capable of doing it."
