Revell grabbed a double and despite a late reply from Andy Robinson for the hosts it was not enough to stop the Millers extending their unbeaten run to four games.
Tranmere came into the midweek clash looking to secure successive home wins for the first time all season but they were always up against it as the visitors dominated throughout.
Wes Thomas thought he had put Rotherham in front after 13 minutes only to be judged offside.
Owain Fon Williams parried James Tavernier's stinging 25-yard drive before Revell converted Kieran Agard's low 39th-minute cross to open the scoring.
Rovers striker Ryan Lowe saw his touch past Adam Collin cleared off the line at the other end but Rotherham were still on top and Thomas missed a sitter immediately after the break.
Agard then saw another shot turned away but Revell smashed home his second goal in the 65th minute from the edge of the box before Robinson pulled one back for Tranmere with a final-minute header.
